Explanation

In the Bible, God forbids Adam and Eve from eating fruit from a tree, which would give the knowledge of good and evil. The Christian interpretation claims that eating the fruit, an event called "fall of man", was a major sin, in fact the first and biggest sin ever committed, because the fruit made humans capable of evil in addition to good. (Note that in the comic, Eve is committing the evil acts of homicide and cannibalism before she has fallen.)

Zach implies the eating of the fruit to have been an intended outcome after all (even if a plan B) to get Adam and Eve to learn about good and evil through reverse psychology. It is well-known that humans hate being told what to do (though evidently their very creator didn't realize this at first): To persuade an immature person, such as someone without knowledge of good and evil, into committing act A, it is often good to command said person to commit act B, which is in essence the opposite of act A, making the person spitefully commit act A as intended all along. This is how God manipulates Eve (and Adam) into eating from the tree, as he specifically commands them to not eat from it.

In addition to contrarianism and spite, Eve displays hubris and pride as she tells God that "nobody tells humans what to do".

The votey subverts the very common belief that God loves humanity.

[Eve is quite clearly eating Adam, covered in blood, and yelling]
Eve: You don't tell me what to do! Nobody tells humans what to do! Nobody!

[Eve is looking at God. There is a tree with fruit.]
God: Eve, I have only one rule. Don't eat from... uh... that tree.
